摘要: Biodegradable polymers are in great demand in tissue engineering. Chitosan is one of the promising natural biopolymers to mimic extracellular matrix (ECM) for tissue repair and regeneration. Currently, porous chitosan matrices have been fabricated using freezing-dry technique and characterized by Scanning Electronic Microscopy (SEM). However, SEM only shows the surface structure of dried materials. It is of paramount importance to develop a method to reveal the real network structure inside the scaffolds. In this study, Fluorescein isothiocyanate (FITC)-labeled chitosan matrices were fabricated and characterized using Confocal Laser Scanning Microscopy (CLSM). The effects of molecular weight and concentration of chitosan as well as the freezing temperature on the three-dimensional (3-D) structure of chitosan matrices were investigated. The real structure of the chitosan matrix was revealed by CLSM without any treatment, which was composed of a network with open-pores interacted in 3-D. The pose size decreased with an increase in molecular weight or concentration of chitosan, and with a decrease in freezing temperature. The structure of matrices showed a fiber-like orientation at low concentration and at -20°C, while it was mesh-like at high concentration or at -80°C. The CLSM provides a useful tool for the study of the structure of chitosan matrices and the interaction of cells with chitosan matrices.
