摘要: Metabonomics, defined as the quantitative measurement of the dynamic multi-parametric metabolic response of living systems to pathophysiological stimuli or genetic modification [ 1 ], has recently demonstrated enormous potentials in many fields such as plant genotype discrimination [2-3], toxicological mechanisms, disease processes, and drug discovery [4-10]. According to the destination, the metabonomics research could be divided into four approaches metabolite target analysis, metabolite profiling, metabolomics and metabolic fingerprinting[11]. The technique platform of the metabonomics was composed of two parts: the platform of the data acquisition and the platform of data interpretation. It is generally accepted that a single analytical technique will not provide sufficient visualization of the metabolome and, therefore, multiple technologies are needed for a comprehensive view. Accordingly, many analytical technologies have been enlisted to profile the metabonome including IR [12], NMR [13-16], TLC [17], HPLC/UV/PDA [18], CE/UV [19], CE/LIF [20], CE/MS [21], GC/MS [2], LC/MS[22], LC/MS/MS [22], FTMS [23], and HPLC/NMR/MS [24]. The bio-informatics methods employed in the metabonomics include two classes of methods [25]: unsupervised method such as Principal Components Analysis (PCA), Nonlinear Mapping procedures (NLM) or Hierarchical Cluster Analysis (HCA) and supervised method that is used to construct mathematical boundaries round each class and to maximize the separation between classes. 2. Establishment of Chromatography-based metabonomics technique platforms Among the analytical technologies mentioned above, NMR is the most popular method used in the metabonomics research, which could provide the high selectivity for all compounds J.K. Nicholson's group has done a lot of the representative work [13-16,24]. While being very effective, NMR has two significant disadvantages: poor sensitivity and resolution, which led to the masking of low abundance analytes by high concentration components. Another popular technology used in the metabonomics research is GC/MS, which could provide higher resolution and higher sensitive compared to NMR. Oliver Fiehn accomplished his work mainly in this platform [2]. But the GC/MS has also its limitation. It couldn't analyze the non-volatile components of the metabolites in the bio-fluids and tissue directly. While most of the metabolites in the bio-fluids and tissues belong to this class. And another limitation is that the peak capacity of GC/MS is still insufficient for the analysis of the all metabolites in the metabonomics.
