摘要: Two classes of sponges(Porifera),the Demospongiae and the Hexactinellida ,can use sillca for the formation of their sillcious skeleton ,called apicules.The underlying biofabrication process sillca structures in sponges is yet largely unknown ,though progresses at both cellular and molecular level have been accumulated in recent years.The current study is aimed to elucidate the biofabrication process of sillca spicules using a newly established in vitro primmorph culture system form archaeocyte-dominant cell population (ADCP)in our laboratory .A sponge Hymeniacidon Perleve collected form China Yellow Sea was used as a model system for the investigation of cell proliferation and in vitro biofabrication of spicules . ADCP was enriched by Ficoll gradient ,which was used for primmorpoh formation .Archaeocyte cells are the totipotent cells among various sponge cell types ,which have been proven to contribute significantly for the imprvenents in the DNA synthesis ,cell proliferation and metablism in primmorph culture .A time –dependent spiculogenesis was clearly demonstrated in our long-term culture ,indicating high metabolic activity .of primmorphs form the ADCP. We further established a flow-through culture system with a time-lapse video microscopy for on-line inverstigation of the biofabrication process of sillca spicules .Date are presented on the cell movements ,spicule formation and self-assembly of primmorpha during in vitor cultivation ,A scheme of sponge cell communication and spicule formation is porposed (Supported by grants form the “Hi-Tech Research and Development Progarm of China ”(2001AA620404)and the European Commission “SILIBIOTEC”)
