题名: Silica Spicule Dvevlopment in in vitro culture of Archaeocyte-Dominant Primmorphs of Marine Hymeniacidon perleve

摘要: sponges(porifera) are the phylogenetically oldest metazoan ,with two calsses ,the Demosepongiae and the Hexactinellida that can use sillica for the formation of their silliceous skeleton ,called spicules .We have recently developed a new method to establish in vitro primmorph culture form archeocyte -dominant cell population (ADCP) enriched by Ficoll gardient ,rather than a mixed cell population (MCP) of sponge Hymeniacidon perleve .The rationale is based upon the totipotency of archaeocyte cells and the different biological funcyions of various sponge cell types .A sponge Hymeniacidon perleve collected form China Yellow Sea was used as a modol system for the investigation of cell proliferation and in vitor evolution of spicules .Distinct dynamics of primmorph formation were observed while significant increases in DNA synthesis ,cell proliferation (up to 3-fold )and cell growth (up to -fold)were achieved .These results demonstrated the importance of archaeocyte and archaeocyte -like cells in successful establishment of in vitor sponge primmorph culture system . Most interesting ,a time -dependent spiculogenesIs was clearly demonstrated in our longterm culture ,indicating high metabolic activity of the ADCP primmorphs .Further date are presented on the cell movements ,spicule formation and self-assembly of primmorphs during in vitro cultivation by a time-lapse video microscopic observation .A scheme of sponge cell communication and spicule formation is proposed .The new in vitro sponge cell culture system is of significant to fundamentally understand the underlying mechanisms for biofabrication of sillca nanostructure materials.
