摘要Abstract:Ginsenoside Rb3, one of protopanaxadiol-type ginsenoside, can be hydrolyzed into Mx, one less - polar ginsenoside by SE. Meanwhile, Mx could be a candidate for exploiting an anti-tumor drug. A ginsenoside-Rb3 hydrolyzingβ-glucosidase was purified from SE through following steps, a)DEAE Sepharose CL-6B ion exchange chromatography (IEC), b) TOYOPEARL Butyl-650C hydrophobic interact chromatography (HIC), c) Sephadex G-200 gel filtration chromatography (GFC).The purified emzyme showed a single band on SDS polyacrylamide gel electrophoresis. The molecular weight was estimated by SDS-PAGE and by gel filtration as 58KD and 59KD,respectivly.The optimal reaction conditions for the enzyme were pH 4.8 at 60℃.Among all tested substrates ,the enzyme exhibited the specific activity to p-nitrophenyl β-D-glucopyranoside(pNPG), also poor hydrolyzing activity to p-nitrophenyl-β-D-cellobioside and p-nitrophenyl α-D-xylopyranoside.
