题名: Effects of medroxyprogesterone acetate on human cytochrome P450 enzymes

摘要: Medroxyprogesterone acetate (MPA), frequently used in contraception and chemotherapy, was reported involving in drug-drug interaction (DDI). However, the mechanism is still unclear. To elucidate whether MPA can cause DDI through the influence on cytochrome P450 enzymes, we conducted the in vitro study of MPA on cytochrome P450 enzymes including CYP1A2, CYP2A6, CYP2C9, CYP2D6, CYP2E1 and CYP3A4. MPA was found to strongly inhibit CYP2C9 and CYP3A4 with an IC50 of 16.1 μM and 31.5μM, respectively, and also slightly inhibit CYP2A6 with an IC50 of more than 100 μM. However, MPA exhibited a stimulation rather than inhibition of CYP2E1. There are no effects observed on CYP1A2. Further study showed that the inhibition on CYPs is not mechanism-based.
