题名: Inhibition of multiple herbs on cytochrome P450 reductase

摘要: Increasing attention is being paid to the inhibition of multiple herbs on cytochrome P450 isoforms (P450) activities, however, their effects on cytochrome P450 reductase (CPR), the electron donator in P450 catalytic cycle, have still not be paid deserved attention to, though the fluctuation of CPR activity might result in a variation of P450 metabolic activity. In this study, some herbs were studied about their effects on six major P450 enzymes and CPR. The herbs are Rhizoma Chuanxiong, Radix Salviae Miltiorrhiae, Flos Carthami, Radix Paeoniae Rubra, Lignum Dalbergiae Odoriferae, Bupleuri, Fructus Aurantii, Radix Notoginseng, Rhizoma Acori Talarinowii, Fructus Aurantii Immaturus, Ramulus Cinnamomi, Spine of Chinese honeylocust, Semen ziziphi spinosae, Xuefu, Guanxin, and Corydalis tuber. The CPR activity was indicated by its specific reaction, MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) oxidation. Their inhibitory effects on CYP3A4, CYP1A2, CYP2E1, CYP2A6, CYP2C9 and CYP2D6 were assayed by HPLC analysis respectively. At 100μg/ml, Miltiorrhiae and Odoriferae exhibited the most potent inhibition of multiple P450 enzymes. Among them, the residue activity of CYP2A6 was 22% by the inhibition of Miltiorrhiae, and the residue activities of CYP3A4, CYP1A2 and CYP2A6 was 10.17%, 3.23% and 14.17% by the inhibition of Odoriferae, respectively. In addition, Rubra and Guanxin inhibited the CPR activity by 20% and 10% respectively. The crude components of Rubra were further separated by resin absorption and extraction of ethanol and n-butanol. The results demonstrated the major inhibitive components of Rubra existed in ethanol eluted solution followed the separation of resin absorption, which caused the reduction of CPR activity by 30%. The inhibition of Rubra to CPR activity might give rise to the inhibitory effects on P450 enzyme activities or cause the occurrence of inhibition on P450 enzymes. Moreover, the inhibitive components of Rubra existed mostly in ethanol eluted solution followed the separation of resin absorption, which could be further separated in the following steps.
