AbstractDeveloping novel HPLC separation materials is one of the most important approaches to resolve the emerging separation problems aroused by the complex samples. The two important aspects of developing HPLC separation materials are 1) to find highly selective and effective reactions for immobilization of the functionalized molecules on the solid support; 2) to design and preparation of functional molecules as novel HPLC stationary phases. The traditional immobilization processes mainly rely on the nucleophilic or electrophilic reactions to obtain the conjugation of solid supports and the functionalized molecules. In this paper, a novel solid/functionality conjugation strategy via “click chemistry” for preparation of separation materials was developed and was proven to be very suitable for preparation of separation materials. Based on the relationship between the structure of bonded functionalities and the chromatographic separation functions, serials of separation materials with novel bonded functionalities were designed. A kind of oligomeric ethylene glycol (OEG) based separation materials for RP-HPLC was designed and prepared. The chromatographic studies demonstrate the OEG columns have universal selectivity for most of the organic compounds in RP-HPLC mode. Furthermore, the OEG columns have more excellent orthogonality with ODS than any kinds of commercial available columns. Novel separation materials with hydroxyl, saccharides and cyclodextrin as functionalities for HILIC were also designed and prepared in our study. The chromatographic results prove these novel separation materials demonstrate well performance in HILIC mode. The applications of these separation materials for separation of Traditional Chinese Medicine were also demonstrated detailedly in this paper.
