题名: “Click oligomeric ethylene glycol”: complementary stationary phases for RP-HPLC

摘要: As we known, RP-HPLC with ODS column is the most popular and efficient chromatographic mode because of the universal selectivity for most of the organic compounds, widely stationary phase supports, simple aqueous mobile phase, etc. However, the traditional separation materials for RP-HPLC face urgent challenges due to the simplex hydrophobic interaction and much more complex samples. Moreover, 2D-HPLC system requires orthogonality between the two columns. Therefore, it is desirable to develop complementary separation materials for RP-HPLC combined with C18. Except for the basic requirement of the chromatographic separation materials, the complementary separation materials for RP-HPLC require universal selectivity, multiple interaction, orthogonality with ODS, etc. In this paper, a kind of oligomeric ethylene glycol (OEG) based separation materials for RP-HPLC was designed and prepared via click chemistry. OEG have similar molecular structure (a flexible chain) and have similar chain length to ODS. Moreover, OEG can provide both hydrophobic interaction and hydrogen bond. The chromatographic studies demonstrate the OEG columns also have universal selectivity for most of the organic compounds in RP-HPLC mode. Furthermore, the OEG columns have more excellent orthogonality with ODS than any kinds of commercial available columns. By the way, the OEG columns have “clustering” function for homogenous component in Traditional Chinese Medicine. The application of OEG column for separation of Traditional Chinese Medicine and for developing methodology of separation techniques are undergoing in our laboratory.
