Two-Dimensional Array HPLC Coupled to Mass Spectrometry: A Powerful High-Throughput Technology to Comprehensive Analysis of the Complex Samples

AbstractAs the realization of the potential important roles that the low-abundant components in the complex samples play, the comprehensive analysis of the complex samples (such as biological samples, environmental samples, natural herbal medicines, etc) is becoming one of the hotspots and difficulties for current analytical chemistry. Multidimensional chromatography is well-known for its powerful separation ability. In comprehensive 2D-HPLC, the analysis speed of the second dimension should be fast enough in order to guarantee completing the analysis before the next sample injection, but the separation ability has been limited by the shorter separation time of the second dimension. In order to enhance the separation ability of LC×LC, longer columns have to be used in the second dimension, but it will lead to longer analysis time. Hence, the balance between the higher separation capacity and faster analysis speed has to be taken into consideration. Inspired by Capillary Array Electrophoresis (CAE), a novel high-throughput method which named 2D-Array-HPLC/MS was developed. It focuses on improving the separation capacity without prolonging the analysis time, and could provide a brand-new way for the development of 2D-HPLC. Longer-columns array were running synchronously in the second dimension, and following the high-resolution 2D chromatographic separation, a multi-channel detector was used. The introduction of “MUX” (multiplexed technology) makes the system simple and cost-effective. In order to show the feasibility of 2D-Array-HPLC/MS, we implement the offline four-channel experiment on the chloroform extracts of Swertia franchetiana Smith, which is a traditional Tibetan medicine for treatment of hepatitis and cholecystitis. 118 compounds can be detected in the chloroform extracts of the plant by the 2D-Array-HPLC/MS method, while there are only 20 well-separated peaks can be found in the one-dimensional chromatogram. Most of the additional compounds we detected were low-abundant components. The peak capacity of the system could be as high as 24300 within 250min at present. Obviously, it could provide a powerful technique for any scientists in the research of low-abundant components in complex samples.
