AbstractUltra high performance liquid chromatography (UPLC) employs 1.7 μm porous particles resulting in higher peak capacity, better resolution and sensitivity compared with traditional HPLC, and has been applied in many researches. Time-of-flight mass spectrometry (TOF-MS), allows the generation of mass information with greater accuracy and precision. These accurately measured mass values can then be used to produce candidate empirical formulae which significantly reduce the number of possible structures of putative compounds. The application of this approach in separation and identification of soybean soponins hydrolyzed by Aspergillus sp.848s was studied in this paper. The enzymatic hydrolysis of soybean saponins sugar-moiety to produce new saponins containing low sugar and high activity was studied. After soybean saponins hydrolyzed by Aspergillus sp.848s, the new saponins were obtained by silica of 1 mm. The optimal developing solvent was chloroform: methanol: water = 74:26:10. The new saponins were analyzed by UPLC/TOF-MS. Hypersil-ODS column (2.1×10 cm, 1.7 μm) was used. Water containing 0.1% HAC: methanol (30: 70) was used as mobile phase, and the velocity of flow was 0.2 ml/min. The temperature of column was maintained at 30 . MSD conditions were that the ℃ion source was ESI, the electro spray needle was held at 3 kV, the temperature and velocity of air was 300 and 500 ml/min, respectively; the scan mode was positive mode and the scan range was 200~1200 aum.℃ In result, the structure of two new saponins was confirmed, which are 3-O-[α-L- arabinopyranosyl] soyasapogenol B and 3-O-[α-L-(1-2)-α-L- arabinopyranosyl] soyasapogenol B. And the molecular weight is 590 and 722 respectively. These results were very useful to give the new more biologic active material for the health food and medicine. The biologic activities of the new soybean saponins need further investigation.
