题名: Studies on pathway of biocatalysis producting terephthalic acid by co-metabolism

摘要: Biological processes are commonly perceived as being less harmful to environment than chemical processes. A variety of chemical routes to synthesize terephthalic acid are known. But biological process to produce terephthalic acid is almost not. The pathway of biocatalysis producting terephthalic acid by co-metabolism of Comamonas testosterone strain DSM6577 and Stenotrophomonas maltophilia was studied in this thesis. A detecting method of middle metabolites produced at the process of transforming p-xylene to terephthalic acid by HPLC was established. The Hypersil SAX column ( 250 mm ×416 mm ×5μm) was used . The temperature of column was 30°C and detected wavelength at 254 nm. The mobile phase was 0.25 mol/L NH4H2PO4 containing 10 % acetonitrile. The flow rate was 0.8 ml/ min. Comamonas testosterone strain DSM6577 was shown to be capable of oxidizing p-toluic acid and stenotrophomonas maltophilia was shown to be capable of oxidizing p-xylene as sole souce of carbon into terephthalic acid through sequential reactions catalyzed by a series of enzymes including monooxygenase and dehydrogenase. P-toluic acid, p-xylene’s degradation and erephthalic acid production were studied respectively. The terephthalic acid producing organism comprises the genes encoding enzymes involved in the enzymatic conversion of p-xylene to terephthalic acid as follows: p-xylene was acted by xylene monooygenase to form 4-methylbenzyl alcohol, which was acted by methylbenzyl alcohol dehydrogenase to form p-tolualdehyde. And then, at the action of tolualdehyde dehydrogenase, p-tolualdehyde was transformed to p-toluic acid, which is acted by toluate methyl monooxygenase to form 4-carboxybenzyl alcohol. At last, 4-carboxybenzyl alcohol was acted by carboxybenzyl alcohol methyl dehydrogenase to form 4-carboxybenzaldehyde, which was acted by caboxybenzaldehyde methyl dehydrogenase to form terephthalic acid. This enzymatic pathway is referred to be the biosynthetic pathway of terephthalic acid.
