题名: Docosahexaenoic acid effects on the disruption of epithelial barrier function through altering lipid composition in membrane microdomains of tight junction

摘要: Recently tight junction (TJ) are considered as raft-like membrane compartments and represent a structural and functional distinct membrane microdomains, which are essential in spatial organization of TJs and in regulation of paracellular permeability in epithelial cells. Flaws in tight junction (TJ) barrier have been considered as a crucial etiologic factor of Crohn’s disease, which is an autoimmune disease and terribly harm human’s health. Polyunsaturated fatty acids, including docosahexaenoic acid (DHA, 22:6 n-3), modulate immune responses and exert beneficial effects on inflammatory bowel disorders. However, the mechanisms are not yet elucidated. In this work, membrane microdomains of TJs were isolated, and raft phospholipids, including Phosphatidyl-ethanolamine (PE), Phosphatidylinositol (PI), Phosphatidylserine (PS) and Phosphatidylcholine (PC), were separated and determined by HPLC-ESI-Qtrap-MS. Then the structures of above phospholipids were identified by HPLC-ESI-MS-MS. Ultrastructure of TJ was observed by transmission electron microscopy. DHA changed phospholipids environment in the membrane microdomains of TJs in vitro. It revealed the effects of DHA on the disruption of epithelial barrier function through altering lipid composition in membrane microdomains of TJs. Moreover DHA could prevent distortion of TJ morphology induced by the cytokines.
