题名: The application of UPLC-QTOF-MS in the metabonomic investigation of hepatocirrhosis

摘要: LC/MS technique has become an important platform and also been widely used in the area of metabonomics. The “Ultra Performance” liquid chromatography (UPLC) employs alkyl-bonded porous particles with a particle size less than 2 μm in diameter operated with increased mobile phase linear velocities thus a significant increase in the resolution results can be gained, such as a better signal-to-noise ratio, increased sensitivity and faster analysis. Because of its sensitivity and resolution, UPLC is applied to the research of metabonomics widely. Here an integrated approach for human serum metabonomic research based on UPLC-QTOF-MS is presented. Totally 30 patients with their diagnosis of hepatocirrhosis and 30 healthy volunteers were enrolled in this study. The chromatographic separation was performed on a 10 cm × 2.1 mm Waters HSS 1.8 μm C18 column, This HSS (High Strength Silica) columns are designed to retain and separate small, water-soluble polar organic compounds. And a 10 cm × 2.1 mm Waters 1.7 μm C18 column was used for a comparison. Mass spectrometry was performed on a Waters Q-TOF micro MS. The raw data were analyzed by the Micromass MarkerLynx,then exported and analyzed by Principal Components Analysis and Partial least squares discriminate analysis using SIMCA-P 11.0. According to the analysis, we found and identified about 10 potential biomarkers from the dataset acquired through the C18 column, including lysophosphatidyl choline, phenylalanine, bile acid, etc. However, 4 new potential biomarkers through the HSS column including glutamic acid, methionine, tyrosine, and valinine. Obviously, the HSS column shows its ability to separate these water-soluble polar organic compounds like amino acids. The integrated metabolites information acquired from different columns enable us to find more potential biomarkers in the metabonomic research of disease. In spite of the wide variability and flexibility of the metabolites in the serum, the method provides us an attractive and powerful tool for the metabonomic research.
