AbstractNow the researches on alkalophilic microorganisms mainly focus on the culture and enzyme extraction and there is few investigation of metabolic pathway. In this paper, Lp3-3 was selected as the experiment strain. The detecting method of metabolites in cell and fermentation liquid was developed based on RP-HPLC, and the influence of environment to metabolism was studied. A detecting method of organic acids in fermentation liquid of Lp3-3 by RP-HPLC was established. The Sinochrom ODS–BP column (250 mm ×4.6mm, 5μm) was used. Mobile phase was 2.5 mmol/L NH4H2PO4 (1% methanol,pH 2.5). The flow rate was 0.5 mL / min, the temperature of column was 30°C and detected wavelength at 210 nm. The method was used to analyze content of organic acids in fermentation liquid of alkalophilic microorganisms, the diversification trend of lactic acid, acetic acid and pyruvic acid at different fermentation time was obtained. In order to study the metabolic pathway of alkalophilic microorganisms, a detecting method of 17 amino acids in cells of alkalophilic microorganisms by HPLC was studied. The chromatographic Sinochrom ODS–BP column was used. Mobile phase were acetonitrile: water=1:1 (solution A) and 0.06mol·/ L + 0.1% N, N – dimethyl – Formamide + 10% acetonitrile, pH was adjusted to 6.4 with 5% acetum (solution B). The flow rate was 0.8 mL / min, the temperature of column was 35°C and detected wavelength at 360 nm. The method was used to analyze content of amino acids in cells of alkalophilic microorganisms at different ferment time, the diversification trend was obtained, and the interrelated metabolizing pathway was studied. By changing culture conditions, the influence of metabolic pathway of Lp3-3 was analyzed, which help us to know the lactic acid dehydrogenase is the main factor to influence the metabolism of Lp3-3.
