Major metabolic traits of rheumatic arthritis patients depicted through genomics and metabonomics

英文摘要To understand biology at system level, examining the structure and dynamic changes under different status can gain more insights than characterizing isolated pathways of cells or organisms. Rheumatoid arthritis (RA), a disease involved multiple organ and system disorders, is still attractive to many researchers and leaves much enigma to be cracked. In this study, we demonstrated the application of the combined genomics and metabonomics strategy to explore the major metabolic traits of RA patients. Peripheral blood mononuclear cells (PBMCs) were isolated from normal controls and the patients. mRNA expression analysis showed 147 differentially expressed genes nearly involved in all biological processes after retrieved through KEGG, Biocarta, SwissPort and Gene Ontology: Biological Processes databases. Sera samples were then subjected to metabonomics assay utilizing ultra performance liquid chromatography- time of flight mass spectrometry. The major metabolites fluctuated statistically significant between the two groups were tryptophan, phenylalanine, and some unsaturated phospholipids. After major serumal protein profiling, concentration differences of albumin, cholesterol, low density lipoprotein (LDL), apolipoprotein A1 and α- hydroxybutyrate dehydrogenase were shared different levels between two populations. Data from identified genes, metabolites and proteins were subjected to correlation analysis; parameters closely correlated were screened out to generate interaction networks for both. Except the components with statistic meanings, vitamin B6 and adrenalin were found to have different correlation scales with others serumal compounds in the two groups. Taken together, these results indicate that this integrative omics driven strategy is a powerful tool for rapid identification of potential markers and key components of RA, and facilitate to reveal the major metabolic changes under disease status.
