摘要: Tocharacterizetheformationofsilicaspicules,thedynamicsofspiculogenesisofanintertidalmarinespongeHymeniacidonperlevis(Montagu1818)(Porifera:Demospongiae)wereinvestigatedbymeasuringthegeneexpressionofsilicatein(theenzymeresponsibleforspiculesilicification)andthedimensionalchangesofspiculesduringthedevelopmentalprocessofindividualspongesandincellculturesofprimmorphsofarchaeocyte-dominantcellpopulations.Thedifferentdevelopmentalstagesofspiculesweredocumentedbytime-lapsemicroscopyandobservedbytransmissionelectronmicroscopyduringa1-monthcultureperiod.Duringitsannuallifecycle,H.perlevishasfourdifferentdevelopmentalstages: dormancy,resuscitation,bloom,anddecline.Field-grownindividualspongesamplesatdifferentstageswerecollectedover7months(MarchtoSeptember2005).Thedimensionsofthesilicaspiculesfromthesesamplesweremicroscop¬icallymeasuredandstatisticallyanalyzed.Thisanalysisandthematerialpropertiesofthespiculesallowedthemtobeclassifiedintofourgroupsrepresentingthedifferentdevelopmentalstagesofspiculogenesis.Silicateinexpres¬sioninthebloomstagewasmorethan100timeshigherthanthatintheotherstagesandwascorrelatedwiththespiculedevelopmentalstage.Thetrendofspiculeformationinfield-grownspongeswasconsistentwiththetrendincellculture.Anewparameter,thematurationdegree(MD)ofspicules(definedastheratioofactualtotheoreticalsilicadepositionofmaturespicules),wasintroducedtoquantifyspiculedevelopment.SilicaspiculogenesisduringH.perlevisdevelopmentwasdelineatedbycomparingMDandsilicateinexpression.
