题名: Molecular Cloning and Characterization of an Oligochitosan induced MAP Kinase from Oilseed

摘要: We isolated a new MAPK cDNA, designated BnMPK4, from oilseed (Brassica Napus H) based on our former microarray data. BnMPK4 encodes a 363-amino-acid protein with high nucleotide sequence similarity to previously reported plant MAPK genes such as AtMPK4. Used RT-PCR technique we found that BnMPK4 has a constitutive expression in seedling leaves and is further up-regulated upon wounding by cut, treatment with global signal jasmonic acid (JA), fungal elicitor chitosan and oligochitosan but don’t respond to salicylic acid (SA). A 69kD GST-BnMPK4 fusion protein was purified by using PGEX-4T-1 vector and antiserum was prepared use this fusion protein. Western blot results showed that BnMPK4 is activated by oligochitosan rapidly. These results suggest a role for BnMPK4 in JA-regulated defense signalling pathway.
