Disruption of marine green alga Platymonas subcordiformis for hydrogenase isolation under anaerobic environment

英文摘要A marine unicellular green alga, Platymonas subcordiformis, was found to evolve hydrogen via photolysis of seawater (Guan et al., 2004). To isolate and purify hydrogenase from P. subcordiformis cells, the protocol for algal cell disruption must be operated under anaerobic environment. To maximize the hydrogenase activity recovery, effect of different cell disruption techniques was investigated including ultrasonication, bead-milling, freeze-thawing and chemosmosis methods. Ultrasonication released the highest concentration of total protein at 2.73mg/mL under the condition of 400W output power, 5 s sonication time, and 10 s interval for 20 times. The yield of hydrogenase (1.44 U/g fresh cell and 1.27 U/g fresh cell, respectively) is higher when the cells were disrupted by glass beads or 0.2 % (v/v) Triton X-100. The protein preparation is however more pure by Triton X-100 method, which was used for further purification of hydrogenase (Yan et al., 2008).
