题名: 2-Haloacid dehalogenase-producing bacteria diversity isolated from marine sponge Hymeniacidon perlevis

摘要: Dehalogenases are important for the bioremediation of increased pollutions by halogen-containing compounds in the environment. 2-Haloacid dehalogenase catalyzes the dehalogenation of 2-haloalkanoic acids to produce the corresponding 2- hydroxyalkanoic acids, with potential production of optically active 2-hydroxyalkanoic acids and 2-haloalkanoic acids (Swanson, 1999; Fetzner and Lingens, 1994). Micro-organisms capable of producing 2-haloacid dehalogenase have been frequently isolated from the soil, but rarely from the sea (Ricarda et al., 1997; Marchesi and Weightman, 2003). Given the high concentrations of halogen ions in the seawater, it is rational to investigate the dehalogenaseproducing bacteria from marine environment. One of the first challenges encountered is the interference of high chlorine concentration of CL in sea water with earlier reported analytical method (Marchesi andWeightman, 2003). In this study, an efficient HPLC-based method was developed for reliable assay of dehalogenase activity and a pH indicator bromothymol blue was employed to guide the bacteria enrichment and isolation from a marine sponge Hymeniacidon perlevis. A total of 139 strains capable of producing the 2-haloacid dehalogenase have been enriched and isolated on agar with 2-chloropropionic acid as only substrate. The strain DEH138 showed a high dehalogenation activity. The strains DEH66, DEH84, DEH90, and DEH138 can also degrade a range of organohalogen compounds. Some strains such as DEH99, DEH130, and DEH138 are found to be halophilic bacteria. Phylogenetic analysis of the 16S rDNA sequences demonstrated a distinctive diversity from earlier reported strains isolated from the soil (Marchesi and Weightman, 2003; p. 5). The strains DEH116, DEH90, DEH99, and DEH66were belonged to the members of Rhodobacteraceae family and up to our knowledge these strains were the first report on the isolation of dehalogenating bacteria.
