AbstractThe commercial mariculture of Gracilaria lemaneiformis has been leading in China, because of their fast growing ability, higher adaptability, uses as aquaculture-bioremediator and for the higher concentration of agar–agar. Besides food and agar-industry, the abundant availability of cultured algae needs other potential and more prospective markets. Based on the established eco-friendly agar extraction technique (Li et al., 2008), we developed a comprehensive utilization technique for the extraction of agar and important proteins fromthe cultured G. lemaneiformis. Through this procedure, the agar production of 19.1%, with the R-phycoerythrin (R-PE) yield of 4.03 mg/g dry alga and the bromoperoxidase (BrPO) yield of 5.12 U/g dry algae were obtained, respectively. The purified R-PE showed three absorption peaks at 498, 535, and 565nm and displayed a maximum fluorescence at 580 nm. The SDS-PAGE analysis showed three bands located at 18, 20 and 34 kDa, which corresponded to , and subunit, respectively. The BrPO obtained from G. lemaneiformis was found to be a monomeric enzyme with a relative molecular mass of 66 kDa (Weyand et al., 1999). The agar produced by this technique had many desirable performances in terms of gel strength, gelling temperature, sulfate content and 3,6-anhydro-l-galactose content (Armisen, 1995). In addition, the remainder products left after the comprehensive utilization have the possibility to be used as the trace element feed additive for domestic animals. This technique is eco-friendly to the environment and workers’ health, and reliable to demonstrating. The results from this study present excellent scope for the commercial application of this new comprehensive utilization technique of G. lemaneiformis.
