摘要: Plasma is one of the most commonly used biofluids in metabonomic studies, as it contains a complex metabolite mixture that reflects global changes resulting from system-wide catabolism and anabolism [1]. Previous metabonomic [2, 3] studies on plasma mainly focus on the metabolites with middle and low polarity, although plenty of highly polar metabolites exist since plasma consists of more than 90% water. In this study, we paid attention to the highly polar contents in human plasma, a sample preparation method was established to extract the highly polar compounds and the hydrophilic interaction chromatography (HILIC) coupled to Q-TOF MS detection was used to analysis the sample. Both positive and negative ionization mode were used to increase the coverage of plasma components. Both positive and negative ionization mode were used to increase the coverage of plasma components. The highly polar fraction of plasma was first inject onto the BEH C18 column and analyzed. It can be seen that all of the highly polar contents couldn’t be retained on C18 column and eluted in dead time. But when using HILIC for analysis, the fraction had a good separation on the column.
