题名: Investigation of alkaloids concentration using solid phase extraction

摘要: Solid-phase extraction (SPE) is an effective tool for the fraction concentration and sample clean-up in the analysis of complex samples [1, 2]. In order to concentrate alkaloids from the preparation fraction of traditional Chinese medicine, many SPE columns including polymer (Oasis HLB), phenyl, CN, C18 were investigated in this work. Several important extraction parameters [3], such as sample loading flow, sample concentration, elution condition were evaluated. The recovery demonstrated that Oasis HLB and C18 were better than others. For the basic property of alkaloids, acidic elution solution was suitable for the high recovery, especially for the C18 SPE column. It can be deduced from the comparison of Oasis HLB and C18 that the adsorption capacity of C18 was more than Oasis HLB. However, the dead adsorption was occurred on C18. For Oasis HLB, the interaction between the stationary phase and alkaloids was a little weaker than C18. So the alkaloids can be easily eluted from the SPE column and had no dead adsorption. It is demonstrated that Oasis HLB was a suitable alternative for the alkaloids concentration from the preparation fraction of traditional Chinese medicine. References: [1] Jin, Y., Xue, X.
