Chitooligosaccharides bind to HUVEC and block its migration by inhibiting no generation

英文摘要Our previous study has certificated the anti-angiogenesis activity of chitooligosaccharides (COS) (Wu et al., 2008), but the mechanism is not clear. In order to investigate how COS perform its anti-angiogenic function, the COSwas labeled by 2-aminoacridone (2-AMAC) and the binding of COS with human umbilical vein endothelial cells (HUVEC) was detected by Flow Cytometry and fluorescence microscope. As nitric oxide (NO) is involved in angiogenesis (Peeyush and Chandan, 2001), the effect of COS on the NO generation ofHUVECwas measuredby Griessmethod and the effect of COS on the actin polymerization of HUVEC was observed under Laser confocal microscopy at the same time. The result showed the binding rate of COS with HUVEC was about 36% and the COS could enter HUVEC and gather around the nucleus. The NO generation assay showed 50 g/ml COS could reduce the NO generation evidently compared to control cells, at the same time 50 g/ml COS could induce the formation of central microfilaments and reduce the number of lamelipodia, compared with what was observed in control cells (Figure). All of the results indicated that COS could bind to HUVEC and block its migration by inhibiting the NO generation.
