题名: Involvement of N-mediated defense in oligochitosan inducing resistance to tobacco mosaic virus

摘要: Oligochitosanwas applied by spraying it on tobacco leaves for inhibition of tobacco mosaic virus (TMV). The maximum inhibition of TMV by oligochitosan was observed when inoculation occurred at 24 h after spraying 50 g/ml oligochitosan (Zhao et al., 2006). But the mechanism is unknown. The tobacco N gene, a member of the Toll-interleukin 1 homology region/nucleotide binding site/leucine-rich repeat (TIR-NBS-LRR) class of resistance (R) gene, confers resistance to tobacco mosaic virus (TMV) (Liu et al., 2004). In this report, we investigated the roles of some genes involved in N signaling, after oligchitosan treating tobacco plants. We used RT-PCR to identify changes of these known defense genes. It was found oligochitosan induced SGT1 and EDS1 gene expression, any of which is required for N function. A mitogen-activated protein kinase (MAPK) OIPK, or an MAPK kinase (MAPKK) NtMEK2, was up-regulated by oligochitosan. Onemember of WRKY transcription factors, TIZZ, which is concerned with N-mediated lesion forming was also induced by oligochitosan. Silencing of the OIPK gene attenuated oligochitosan-inducing resistance to TMV (Feng et al., 2006). We also found silencing OIPK altered oligochitosan-triggering genes expression, and further silenced TIZZ. Our results suggest that oligochitosan inducing tobacco defense is involved with N-mediated resistance.
