AbstractFor shotgun method, protein digestion is one of the most important steps, which is usually performed in solution, but recently more focus is put on the trypsin microreactor because of the high digestion efficiency. However, in most cases, the pH and organic reagent concentration of protein solutions, especially for eluants from HPLC separation, are not compatible with immobilized trypsin. Therefore, it is indispensable to develop on-line buffer exchange interfaces (1). In our work, a novel online buffer exchange interface based on microdialysis was developed, and successfully applied into the adjustment of pH and acetonitrile concentration successfully within 30 seconds.
