题名: Cationic micelles prepared by amphiphilic chitosan derivatives for siRNA delivery

摘要: Abastract In the last decade, a lot of studies on preparation of varied nanocarriers with cationic polymers such as PEI, PLL, chitosan and their derivatives have been reported for DNA and siRNA delivery [1]. Besides the familiar nanoparticles, cationic micelles have also been utilized to deliver siRNA and DNA recently [2-5]. Here, we present the design and synthesis of new amphiphilic chitosan (CTS) derivatives, and then the preparation and characterization of cationic micells for the potential application on loading, delivering and releasing of siRNA. In this study, LA, a kind of fatty acid that is a metabolite in human body, was firstly conjugated to CTS to get amphiphilc chitosan-LA (CTS-LA). Subsequently, the CTS-LA was quaternized to get N-trimethyl chitosan-LA derivatives (CTS-LA-TM) with good water solubility in broad pH range. A serial of CTS-LA-TMs were synthesized with different degree of substitution (DS) of LA (2.0-3.0%) and different degree of quaternization (DQ) rangeing from 40% to 70%, which was characterized with FTIR, elemental analysis, 1HNMR, and XRD. Self-assembly of these amphiphilic polymers into cationic micells in aquoues solution was also studied by fluorescence techniques. These micelles fabricatd at pH 7.4 (0.02M) showed critical micelle concentration (CMC) varied from 0.04 to 0.01mg/mL, and the average size rangeing from 130 to 210 nm with zeta potential around 20mV when the DS of LA increased from 2.0% to 3.0%. Furthermore, zeta potentials of these micelles were enhanced by reducing solution pH, displaying certain DNA or RNA binding ability. Currently, the loading of siRNA into these cationic micelles and in vitro evaluation are undergoing in our lab.
