摘要: Analysis of whole proteome is an extraordinary challenge because of the complexity and wide dynamic range of proteins,Therefore protein prefractionation is pre-requisite to achieve high efficient proteome analysis.In this study gel-free protein fractionation by preparative microscale solution isoelectric focusing(PMSIEF).Off-line protein digestion,peptide separation by 2D-SCX-RPLC with serially coupled long microcolumn, and protein identification by ESI-MS/MS were combined,by which 1852 proteins were identified from rat brain.
