题名: A two-dimensional liquid chromatography (NPLC/RPLC) with vacuum evaporation interface and complete solutes transfer

摘要: A comprehensive two-dimensional liquid chromatographic system incorporating a vacuum-evaporation interface was developed. A Normal-phase liquid chromatography (NPLC) with a CN packed capillary column (200 mm  0.53 mm i.d.) was used as the first dimension (1st-D), and a reversed-phase liquid chromatography (RPLC) with a C18 packed capillary column (150 mm  0.32 mm i.d.) was used as the second dimension (2nd-D). An electronically controlled dual-position, ten-port valve with two identical storage loops served as the interface. The switching time interval was adjustable. The solvent in the loops of the interface was evaporated dynamically at 25 oC under vacuum conditions, leaving the solutes on the inner wall of the loops. The mobile phase of the 2nd-D dissolved the solutes in the loop and injected them onto the second column, performing an on-line solvent removal of the fractions from the 1st-D and change of the mobile phase of the 2nd-D. The chromatographic separation of analytes on the two dimensions was optimized individually without interaction. Sample loss due to evaporation in the interface was investigated with standard samples having different boiling points. It was found that the recoveries of the standards on the proposed system were roughly proportional to their boiling points, and the recoveries of non-volatile compounds were all above 50%. The applicability of the 2-D LC system was demonstrated for the analysis of traditional Chinese medicine extracts such as Radix salviae miltiorrhiza bage and Angelica dahurica. The interface for 2-D LC developed in this work could realize fast solvent evaporation of 1st-D fractions, reduce the injection volume of the 2nd-D, and achieve band compression at the head of the second column. The chromatographic performance of the NPLC/RPLC system was enhanced without sacrificing the separation efficiency and selectivity of each dimension, leading to the advantages of high peak capacity, true orthogonal and high resolution.
