AbstractMonolithic column is a good alternative to particulate column due to its high permeability and mass transfer rate, as well as easiness to preparation. We have developed an online multidimensional separation system using a 7 cm × 150 μm i.d. monolithic strong cation exchange (SCX) capillary trap column. And this system exhibited powerful performance in LC separation and proteins identification [1-2]. This system was successfully applied to investigate the mechanism of lipid accumulation by the oleaginous yeast Rhodosporidium toruloides by label-free quantitative proteome analysis and the differential protein expression in Huntington’s disease by SILAC labeling. We also optimized the preparation and LC separation conditions of the C12 hydrophobic monolithic column to increase the proteome coverage in μLC-MS/MS analysis [3]. We prepared a 7 cm × 25 μm i.d. monolithic ESI emitter to replace the commercial available void capillary emitter. This type of monolithic ESI emitter could increase 12.8% peak capacity of the μLC-MS/MS system as well as improve 130.0% proteome coverage in label free quantitative proteome analysis [4]. A particulate capillary column with integrated monolithic frit was also developed. This type of column can be packed with stationary phase particles with small diameter more easily and would not be clogged during packing and separating process comparing to fritless capillary column. The monolithic emitter integrated on the packed capillary column can improve the quality and efficiency of the electrospray ionization and can be regenerated, which is important for qualitative and quantitative proteome analysis [5].
