AbstractLung cancer is the second most diagnosed but the number one cause of cancer-related death in cancer [1]. Radiation therapy has been believed to be a potent technique in the current cancer therapy. As cancer is a disease related to genomic changes, the disease itself or the treatment of the disease would give rise to changes in physiological systems of lung cancer patients, a systematic approach such as metabonomics would be very helpful to disease diagnose and therapy toxicity research. But to the best of our knowledge, there were no metabonomic studies of lung cancer or the toxicity of radiotherapy on cancer patients. We have previously reported a method to study the highly polar metabolites in the plasma based on ultraperformance hydrophilic interaction liquid chromatography coupled with Q-TOF mass spectrometry [2]. In this study, plasma were collected from 26 health volunteers and 51 lung cancer patients in different radiotherapy periods. The highly polar metabolites in the plasma were extracted,and metabnomic studies based on the developed method in previous experiment were carried out. Difference of these highly polar metabolites in the healthy persons’ and lung cancer patients’ plasma had been studied by multivariate statistical analysis. The result showed a good classification between the health volunteer and the lung cancer patients, and several potential biomarkers were found according to parameter VIP and T test. Moreover, a clear discrimination among the patients in different radiotherapy periods was noted.
