AbstractA type of characteristic peroxisome in the spherulous cells of the marine sponge Axinella sp Debromohymenialdisine (DBH) and hymenialdisine (HD) are two bioactive alkaloids with pharmacological activities for treating osteoarthritis and Alzheimer’s disease. These compounds had been isolated from the marine sponge Axinella sp., and were recently cellular localized particularly in the spherulous cells. Establishing a marker for this type of cells is required for studies on cell differentiation and metabolism regulation of the two alkaloids in in vitro cell culture system. In this research, a type of peroxisome and the relevant peroxidase was detected as markers exclusively in spherulous cells. First, the peroxidase activities in different cell fractions were examined using a cell separation protocol and followed by cytochemical staining with 3,3 -diaminobenzidine (DAB). With the separation protocol, the sponge cells were divided into three fractions, consisted of enriched spherulous cells, the large sponge cells, and the small sponge cells, respectively. The subsequent staining exhibited a distinguishing feature of dark brown granules in cytoplasm exclusively in spherulous cells. Then the transmission electronic microscopy (TEM) was employed to examine the inner structure of the spherulous cells. Two kinds of inclusions were observed inside, of which the larger ones, with a regular oval shape, smooth and even in texture, were found to be the responsible peroxisomes in DAB staining, as they showed consistency in dimension and amount with the positive stained granules observed in light microscopy. Finally, a native–PAGE with protein extracts from different cell fractions was conducted to show the relevant peroxidase. The assay displayed a solely distinctive band with peroxidase activity in the spherulous cell extracts, with a molecular weight of 220kd. This was the first application of DAB staining in sponge and worked out a characteristic peroxisome in spherulous cells. These results would benefit cell identification and also initiate the studies on peroxisome function and their role in DBH/HD metabolism.
