AbstractLycium ruthenicum Murr., is a famous traditional Chinese herbal medicine which widely distributes in salinized desert of Qinghai-Tibet Plateau. Its ripe fruits had been used for treatment of heart disease, abnormal menstruation and menopause. Likewise, polysaccharides isolated from L. ruthenicum were shown to have antidiabetic and antifatigue effects. But up to now no comprehensive study has been conducted to explore the structural characterization of L.ruthenicum polysaccharides. This has largely restricted the further research and development of L. ruthenicum. In this study, the crude polysaccharide was extracted following the usual separation procedure, including water dissolution, centrifugation and precipitation with ethanol. The crude polysaccharide was then applied to anion-exchange chromatographyand gel filtration to obtain a homogeneous polysaccharide, designated as LRGP1. Its molecular weight was 56.2 kDa determined by HPGPC. Monosaccharide composition analysis revealed that it was composed of rhamnose, arabinose, xylose, mannose, glucose, and galactose in a molar ratio of 4:48:1:2.2:3.7:41. LRGP1 had IR bands at 800-1200cm−1, 1450-1800cm−1, 2500-3000cm−1, and 3200-3600cm−1, which were distinctive absorptions of polysaccharides. On the basis of total acid hydrolysis, partial acid hydrolysis, methylation and ESI-MS analysis, LRGP1 was a branched polysaccharide rich in arabinose and galactose with a backbone composed of →6)-Gal-(1→. The branches were composed of (1→5)-linked- Ara, (1→2)-linked-Ara, (1→2,4)-linked-Rha and (1→3)-linked Gal.Arabinose, xylose, mannose, and glucose were located at the terminal of the branches.
