题名: Alginate gel scaffold culture system sa a mimic of the hepatocellular carcinoma microenvironment

摘要: The characteristics of cells cultured within three-dimensional (3D) structures rather than onto two-dimensional (2D) culture plastic more closely reflect their in vivo responses. To elucidate how 3D structure may affect hepatocarcinoma cellular responses, we compared the cancer cell behavior of human liver hepatocellular carcinoma cell line (LM3) cultured as monolayers or as spheroids within alginate gel (ALG) beads. Within ALG beads, cells showed a more malignant in vivo-like phenotype with long-term viability compared to 2D culture. Real-time quantitative PCR analysis showed that the expression of metastasis-related proteins and adhesion-related proteins increased in ALG beads compared to 2D in a time-dependent manner. Furthermore, the expression of genes involved in liver function and self-renew also elevated in the ALG beads. Higher survival rates after exposure to the anti-cancer drug cisplatin were observed in cell spheroids in ALG beads (65%-75%) compared to 2D culture (40%). These results suggest the 3D culture system used in this study has mimicked microenvironment similar to the in vivo performances.
