英文摘要The ordinary method of polymerase chain reaction (PCR) product identification is to let the PCR product comigrate with some standard deoxyribonucleic acid (DNA) fragments, the length of the PCR product can be estimated by the migration time. Undenatured buffer or normal buffer systems were often employed to separate DNA fragments. Because the migration behavior of DNA fragments in the capillary was determined by fragment length as well as conformation, denaturant such as urea should be used in the buffer to eliminate the influence of conformation. The author found that when the PCR products of SRY (sex-determining region of the Y chromosome) and inner contrast 108/B comigrated with DNA standards PBR322/hae III, peak reversion can be noticed. This kind of peak reversion is the result of conformation influence. When estimating the length of the PCR product by calibrating method at this time, false result may be obtained. In a denatured buffer contained urea as denaturant, the PCR products appear at the right place in the electropherogram, which indicates that DNA migrates only according to its fragment length. A more precise result can be obtained when estimating the length of the fragments in a denatured non-gel sieving system.
