AbstractThe matrix of 3, 4-diaminobenzophenone(DABP) was applied in the analysis of phospholipids by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. The optimal composition of DABP matrix solution with methanol-water-hydrochloric acid (80 20 3, V/V) was used in the analysis of different phospholipids species by comparing with the conventional matrices such as 2,4,6-trihydroxyacetophenone(THAP), 2,5-dihydroxy-benzoic acid (DHB) and alpha-cynao-4-hydroxycinnamic acid (CHCA), 3, 4-diamino-benzophenone ( DABP) can effectively suppress alkali metal ion adducts, thus showing single peak of phospholipids with low laser energy and strong peak strength. 500 fmol phosphatidylcholines(PCs) can be detected with high signal to noise ratio. With DABP as a matrix for the analysis of egg yolk and soybean extracts, intact phospholipids ion peaks can be readily produced with clean spectrum background and high resolution. In addition, the sample preparation method is simple and time-saving.
